Initial Steps towards Biocontrol in Hops: Successful Colonization and Plant Growth Promotion by Four Bacterial Biocontrol Agents

چکیده
Verticillium wilt, caused by Verticillium nonalfalfae and V. dahliae, is a devastating disease in hops that can cause considerable economic crop losses. The perennial use of hops combined with the long persistence of the pathogen in soil make it difficult to suppress the disease with conventional measures. Biological control agents (BCA) are the basis of an environmentally friendly plant protection strategy that uses plant promotion and antagonistic effects of microorganisms. We evaluated the effect of four selected beneficial bacterial strains, Burkholderia terricola ZR2-12, Pseudomonas poae RE*1-1-14, Serratia plymuthica 3Re4-18, and Stenotrophomonas rhizophila DSM14405 T for their use in hops. All strains were shown to be both rhizosphere and endorhiza competent, and their abundances ranged from log10 3.0 to log10 6.2 CFU g −1 root fresh weight in the endorhiza and from log10 2.9 to log10 4.7 CFU g −1 root fresh weight in the rhizosphere with B. terricola ZR2-12 showing the highest overall cell densities. Microscopic visualization of DsRed-labeled transformants with confocal laser scanning microscopy showed different colonization patterns and confirmed the rhizosphere competence. Growth promoting effects on seedlings treated with bacteria were found for OPEN ACCESS Agronomy 2013, 3 584 S. plymuthica 3Re4-18 and S. rhizophila DSM14405 T . Competent colonization and plant growth promoting effects are the most important prerequisites towards efficient biocontrol.

Background: Random induced mutation by gamma radiation is one of the genetic manipulation strategies to improve the antagonistic ability of biocontrol agents. Objectives: This study aimed to induce mutants with more sporulation, colonization rate leading to enhanced antagonistic ability (in vitro assay) comparing to wild type (WT) and the assessment of genetic differences (in situ evaluat...
متن کاملBiocontrol activity of endophytic fungus of barley, Microdochium bolleyi, against Gaeumannomyces graminis var. tritici
In this study, two isolates of genus Microdochium (W2 and B26) was isolated from the roots of healthy barley plants in agricultural fields from Kermanshah province in 2014 and were identified as Microdochium bolleyi based on morphological and molecular characteristics. Dual culture studies revealed that W2 and B26 inhibit 45% and 20% of the radial growth of Ggt in tur...
متن کاملBacterial selection for biological control of plant disease: criterion determination and validation
This study aimed to evaluate the biocontrol potential of bacteria isolated from different plant species and soils. The production of compounds related to phytopathogen biocontrol and/or promotion of plant growth in bacterial isolates was evaluated by measuring the production of antimicrobial compounds (ammonia and antibiosis) and hydrolytic enzymes (amylases, lipases, proteases, and chitinases)...
متن کاملDraft Genome Sequence of Chryseobacterium sp. Strain GSE06, a Biocontrol Endophytic Bacterium Isolated from Cucumber (Cucumis sativus)
Chryseobacterium sp. strain GSE06 is a biocontrol endophytic bacterium against the destructive soilborne oomycete Phytophthora capsici, which causes Phytophthora blight of pepper. Here, we present its draft genome sequence, which contains genes related to biocontrol traits, such as colonization, antimicrobial activity, plant growth promotion, and abiotic or biotic stress adaptation.

In this study, Bacillus subtilis 9407 showed a strong antibacterial activity against Acidovorax citrulli in vitro and 61.7% biocontrol efficacy on melon seedlings 4 days post inoculation under greenhouse conditions. To understand the biocontrol mechanism of B. subtilis 9407, identify the primary antibacterial compound and determine its role in controlling bacterial fruit blotch (BFB), a srfAB d...
